Description

JS6 is a Bcl3 inhibitor, and inhibits Bcl3-NF-kB1 binding. JS6 inhibits tumor cell growth in vitro and in vivo.

Biological activity

In Vitro JS6 potently inhibits Bcl3 activity in cancer cells, reducing Bcl3-NF-κB1 binding, tumor colony formation, and cancer cell migration[1]. JS6 (Compound 1a) (24 h) reduces the viability of MCF-10A, MDA-MB-231 and SKBR3 breast cancer cells[2]. JS6 (24 h pre-incubation followed by 24 h chamber incubation) reduces the motility of MDA-MB-231 cells overexpressing Bcl-3[2]. JS6 (24 h) inhibits the binding of Bcl-3 to p50 in HEK-293 cells overexpressing FLAG-tagged Bcl-3[2]. JS6 (pre-incubated for 24 h, followed by 48 h of incubation after transfection) inhibits the NF-κB signaling pathway in Bcl-3-overexpressing MDA-MB-231 cells, Bcl-3-overexpressing HEK-293 cells, and HEK-293 cells overexpressing both Bcl-3 and p52[2].
